چکیده

Background: To determine the effect of combined supplementation of pioglitazone and levo carnitine on plasma glucose, insulin resistance and serum adiponectin in T2DM mice. Methods: In this randomized controlled trial 40 healthy BALB/c mice were divided into four groups (n=10 each). Mice were fed high fat diet for two weeks followed by intraperitoneal injection of streptozotocin to induce T2DM. Group I served as diabetic control. Group II was administered pioglitazone. Group III received levo carnitine and group IV was supplemented combined pioglitazone and levo carnitine. After six days of supplementation, terminal intra-cardiac blood extraction was carried out and samples were analyzed for plasma glucose, insulin resistance and serum adiponectin levels. Results: Plasma glucose levels and insulin resistance significantly decreased in the combined supplementation group compared to the diabetic control. Serum adiponectin levels showed significant increase in combined supplemented group when compared with diabetic control and levo carnitine groups. The data also revealed a positive correlation between plasma glucose and insulin resistance and a negative correlation of these two parameters with serum adiponectin levels. Conclusions: The combined supplementation of pioglitazone and levo carnitine significantly improved glycemic control, insulin resistance and serum adiponectin vis-à-vis individual supplementation in type 2 diabetic mice.

Antihyperglycemic effect of Rosa damascena is mediated by PPAR.γ gene expression in animal model of insulin resistance

Insulin resistance is a condition in which insulin signaling and action are impaired in insulin sensitive tissues and results in hyperglycemia, hyperlipidemia and type 2 diabetes mellitus. Our previous studies have shown that rosa damascena has antihyperglycemic effects on diabetic and normal rats. Therefore, we conducted a study to evaluate the effect of this medicinal plant on insulin sensiti...

Inhibition of Interleukin-1 Receptor-Associated Kinases 1/4, Increases Gene Expression and Serum Level of Adiponectin in Mouse Model of Insulin Resistance

Insulin resistance is a feature of most patients with type 2 diabetes mellitus. Epidemiological evidence suggests a correlation between inflammation and insulin resistant states such as obesity, but the underlying mechanisms are largely unknown. Interleukin-1 receptor-associated kinases (IRAK) play a central role in inflammatory responses by regulating the expression of various inflammatory gen...

The Effect of Co-administration of Pioglitazone and Simvastatin on Insulin Resistance Parameters and PPAR.γ Expression in Insulin-resistant Rats

Backgrounds: Insulin resistance is a pathological condition associated with metabolic syndrome. In this condition, insulin action in liver, muscles, and adipocytes decreases which leads to hyperglycemia, hyperinsulinemia, and dyslipidemia. Thiazolidinediones (Pioglitazone) have been used to enhance insulin sensitivity but due to dyslipidemia associated with insulin resistance, ...
